Degradation

Definition

Degradation is the gradual breakdown or loss of the original properties of a substance, material, molecule or biological system. The term is used throughout science, including pharmaceutical research, biology, chemistry and biotechnology. What happens during degradation varies with the substance and the conditions surrounding it. Degradation is simply when something is not quite the same as it used to be. You might break a molecule into smaller compounds. The material might lose its strength. Enzymes might break down a biological substance. The process is also crucial in the field of research, taking into account that degradation alters the behaviour, duration and appropriateness of a substance.

What causes degradation?

Multiple factors cause degradation. Different materials respond to different environmental and biological factors. Temperature is a shared factor. Some compounds are not stable to long exposure to heat. Temperature-controlled storage is thus important for medicines, laboratory reagents and biological samples. Degradation can also be caused by light. Some molecules are sensitive to ultraviolet or visible light and may change chemically. Another process called photodegradation is involved in the process. A substance may undergo hydrolysis, as it reacts with moisture and water. The rate of the hydrolysis of a specific substance might also depend on the pH, temperature, and its chemical composition. Oxygen may also play a certain role through oxidation, which can modify the structure of molecules and result in unwanted by-products. Substances may also be broken down by enzymes and other natural processes in biological systems. Degradation is usually studied under controlled conditions because there are several factors that may be involved and to understand what causes a substance to change.

Degradation in the pharmaceutical research

Degradation is especially important in pharmaceutical development. A drug must be stable enough during manufacturing, storage and use. If the active element deteriorates quickly, the medicinal product will not function properly. Stability research is used by scientists to find out how a material behaves over time. The samples may be subjected to different environmental factors such as temperature and humidity. Thus, it is possible to analyse samples and ascertain if the original matter still exists and if there are any degradation products formed. The data derived can help find out if the compound has the required storage and shelf life. It can also help identify any possible issues related to the development of drugs. For instance, the compound can preserve its properties at room temperature but will decay faster at higher temperatures. Therefore, its storage, transportation, or formulation must be assessed accordingly. Another aspect is deterioration products. The researchers should know what the original compound changes into while conducting stability studies.

Decomposition of protein and peptide

Peptides and proteins are not easy to degrade due to their structural complexity. Either they are modified or degraded by means of proteases because degradation influences the stability and activity of a drug based on a peptide. A peptide may not work if it is decomposed before it reaches the target. The studies of degradation can be done in different environments, which may be laboratory conditions, biological fluids or experimental models. The results show how the molecule behaves in biological conditions and whether any changes to its structure, formulation or delivery can enhance its stability.

Chemical degradation and biological degradation

It is good to define whether the degradation is chemical or biological because there could be some similarities.

Chemical degradation refers to changes that occur due to chemical reactions. Typical examples are hydrolysis, oxidation and photodegradation. These reactions are able to change the molecular framework of a substance in the absence of a biological system.

The other is a biological degradation, which is a biological process. This process consists of the division of a compound into smaller pieces with the help of enzymes, bacteria, and other living organisms. This aspect could be crucial for the interpretation of scientific findings. A compound may behave normally in a non-biological environment and yet be different in a biological environment where enzymes and other molecules are present.

Why is the degradation study so important?

The significance of degradation studies lies in the practical questions researchers can answer: How can a substance be stable for a certain amount of time? What makes it ineffective? How fast does the process take place? What do the products do? Does degradation affect biological activity? In pharmaceutical work, degradation studies can aid decisions on formulation, packaging and storage. In biological research they can help scientists understand what molecules are doing inside cells or organisms. One can study degradation in environmental science in order to learn about the behaviour of substances or chemicals in the soil, water or other types of environments. Degradation can be advantageous instead of being a bad phenomenon. Certain biodegradable materials are intended to break down when exposed to appropriate biological or environmental conditions.

Degradation does not always mean complete breakdown.

Degradation does not mean the substance is lost completely. It may just turn into something else. The molecule can lose some of its structure and retain some of it. It may produce one or more degradation products, which differ in their chemical or biological properties. Some may be innocent; others will need further investigation. The rate and magnitude of degradation can be as important as whether degradation has occurred.

Degradation in research models

When researchers look at a new compound, they might use different research models to study degradation. Laboratory experiments can provide controlled information about chemical stability. Cell-based systems or animal models may give insight into biological breakdown. The selection of a model depends on the research question. A simple chemical stability test can indicate if a compound is sensitive to heat or water. More complex studies may be needed to understand the effects of enzymes, tissues or whole biological systems. Degradation is therefore often considered together with properties such as absorption, metabolism, stability and biological activity.

In summary

Degradation is basically about change and breakdown. Heat, light, moisture, oxygen, enzymes or other environmental and biological factors may affect a substance. This knowledge of changes can help a researcher in the pharmaceutical and life science research fields to evaluate stability, degradation products and the possible behaviour of a compound over time. The process is not always a negative one. Degradation can be problematic, particularly if it results in the loss of stability or activity of a useful compound. In other cases, controlled degradation is desirable and part of how a material or biological substance is intended to be used. A complete understanding of degradation allows the researcher to get the complete picture of the object being studied.
